The bucket tilt cylinder has a small leak on the top side. The hydraulic hose is brand new. I would like to remove the hose and apply some Teflon tape to the threads. With the engine NOT running, can I simply remove the hose. Will the hydraulic hose be under pressure?

as best as I know they do not suggest teflon tape on hydro connections as it can not take the running pressure, look at the fitting and see if there is a o-ring , some do some dont . most are like a flared fitting (jic), which should seal. if it is a new hose make sure the leak is not coming from under a bad crimp.
